Application Note Cell culture for live cell imaging
This application note presents how to perform cell culture on chip using the Perfusion Pack and the MicroSlides developed by ALine Inc.
This systems uses continuous perfusion, creating a stable environment for the cells, without any manual operations.
The MicroSlides are designed to avoid applying shear stress on your cells. The cells are not directly put into the flow.

How to use the Perfusion Pack
Before starting the experiment, rinse the MicroSlide, the reservoirs and all the tubes and connectors with 70% ethanol to ensure sterility. Please make sure to perform all the following steps under a biosafety hood to avoid contamination.
Fill the reservoir with the culture medium and connect the reservoir to the flow controller.
Connect the reservoir to the MicroSlide.
How to fill the microslide
Once the MicroSlide is connected to the Perfusion Pack, tilt the device as shown on the picture. Using the Elveflow Smart Interface Software, activate the pump until all three wells are ¼ filled then shut off.
Add 10- to 30 uL of sample to each well with a micropipette.
Remove the adhesive liner from the MicroSlide, and seal it with the lid. Use thumb pressure to seal the lid.
