This application note presents how to perform cell culture on chip using the Perfusion Pack and the MicroSlides developed by ALine Inc.

Tip: Medium or drug switch

It is also possible to perform medium switch in order to expose the cells to different drugs or conditions. Check our dedicated application note for this specific setup.

Tip: No more bubble

A bubble trap can be added just before the MicroSlide to make sure that air bubbles don’t enter the chip.

How to use the Perfusion Pack

Long term cell culture during live cell imaging sterility

Before starting the experiment, rinse the MicroSlide, the reservoirs and all the tubes and connectors with 70% ethanol to ensure sterility. Please make sure to perform all the following steps under a biosafety hood to avoid contamination.

Fill the reservoir with the culture medium and connect the reservoir to the flow controller.

Connect the reservoir to the MicroSlide.

Once the MicroSlide is connected to the Perfusion Pack, tilt the device as shown on the picture. Using the Elveflow Smart Interface Software, activate the pump until all three wells are ¼ filled then shut off.

Add 10- to 30 uL of sample to each well with a micropipette.

Remove the adhesive liner from the MicroSlide, and seal it with the lid. Use thumb pressure to seal the lid.

During the experiments, the MicroSlide and the reservoirs can be placed inside an incubator or environmental chamber, while the OB1 and flow sensor stay outside. Use longer pieces of tubing in order to place the instruments outside the incubator.
